Change in the structure of Escherichia coli population and the pattern of virulence genes along a rural aquatic continuum ArchiMer

The aim of this study was to investigate the diversity of the E. coli population, focusing on the occurrence of pathogenic E. coli, in surface water draining a rural catchment. Two sampling campaigns were carried out in similar hydrological conditions (wet period, low flow) along a river continuum, characterized by two opposite density gradients of animals (cattle and wild animals) and human populations. While the abundance of E. coli slightly increased along the river continuum, the abundance of both human and ruminant-associated Bacteroidales markers, as well as the number of E. coli multi-resistant to antibiotics, evidenced a fecal contamination originating from animals at upstream rural sites, and from humans at downstream urban sites. Excretion Products of Shigella dysenteriae and Apoptotic Cell Death on Chick Embryo Muscle Tissue International Journal of Morphology

The aim of this study was to evaluate the acute cell injury of excretion products present in culture filtrate from Shigella dysenteriae in both whole lower limb of chick embryo ex vivo and myoblasts cells developed in hanging-drop cultures in vitro. Three controls were defined: a) Tyrode's solution b) brain-heart broth infusion (CCC) and c) supernatant not toxigenic of E.coli 0157:H7. Shigella dysenteriae were culture for 24 hours and the excretion products were obtained after centrifugation of the culture. Histidine-Tagged Shiga Toxin B Subunit Binding Assay : Simple and Specific Determination of Gb3 Content in Mammalian Cells OAK

A two-step binding assay for globotriaosylceramide (Gb3) content was developed by histidine-tagging strategy, which is a well-established method for the purification of recombinant proteins. The complete binding of the recombinant His-tagged Shiga toxin 1B subunit (1B-His) (1 microg/ml) to the standard Gb3 adsorbed on a multi-well H type plate was observed within 30 min at 37 degrees C; and its binding could be visualized by the following applications of HisProbe-HRP (8 microg/ml) and tetramethylbenzidine (TMB) peroxidase substrate. The 1B-His binding assay was linear over the range of 1 to 100 ng of Gb3 per well. Prevalence and characteristics of Shiga toxin-producing Escherichia coli (STEC) strains in ground beef in São Paulo, Brazil BJM

This study aims to assess the prevalence of Shiga toxin-producing Escherichia coli (STEC) in ground beef collected in two cities located in the State of São Paulo, Brazil. A total of 250 samples of raw ground beef were collected in local grocery stores during the period of March to December 2002 in the cities of Ribeirão Preto (114 samples) and Campinas (136 samples), São Paulo State, Brazil. The samples were processed according to standard methods. The resulting 591 E.coli colonies were screened for STEC by hybridization assays using the specific DNA probes, stx1,stx2 and eae.
